重叠法联合长引物法构建具有标签的突变体

摘要:目的 建立一种快速构建具有标签的突变体的方法。方法 以构建具有HA标签的DJ-1 L166P突变体(DJ-1 L166P-HA)为例。DJ-1隶属于ThiJ/PfpI蛋白质家族,DJ-1 L166P突变导致早发的常染色体隐性遗传的帕金森病。采用重叠法联合长引物法, 克隆DJ-1 L166P-HA至pcDNA3.1(+)。结果 只需一次连接反应,成功地构建了具有HA标签的DJ-1 L166P突变体。该重组质粒转染HEK293细胞后,DJ-1 L166P-HA融合蛋白质得到表达。结论 该方法速度快,易于掌握,较其它方法具有显著优越性。

Cloning of Mutant with Tag by Overlap Method Combined with Long-primer Method
Abstract:Objective Introduce a method which can construct mutant with tag rapidly. Methods Take the construction of DJ-1 L166P mutant with HA as an example. DJ-1 belongs to ThiJ/PfpI protein family, and DJ-1 L166P mutation can lead to autosomal recessive early-onset Parkinson's disease. Using overlap method combined with long-primer method, the DJ-1 L166P-HA was cloned to pcDNA3.1(+). Results We have cloned DJ-1 L166P mutant with HA epitope successfully by one time ligation. After the recombinant plasmid transfects HEK293 cell, the DJ-1 L166P-HA fusion protein can be expressed. Conclusions The method has more advantages than others, for it is easy to be mastered and fast to be conducted.
