茶皂素提取、抑菌活性的研究及应用

摘要:以脱脂后的茶籽饼为原料,采用水提醇萃法和大孔树脂法相结合,并对提取纯化条件进行了优化。采用酶标仪测定了茶皂素对几种常见微生物生长的影响,并测定最小抑菌浓度(MIC);利用浊度法检测茶皂素的热稳定性能;运用正交实验设计法确定洗手液的典型配方,并检测其感官指标,理化指标及微生物指标。结果表明,水提最佳条件:浸提温度70℃、液固比8:1、浸提时间4h。在茶皂素对7种实验菌具有较好的抑菌效果,尤其对细菌有很好的抑制效果,其MIC值分别为:0.5mg/mL、1mg/mL、2mg/mL、2mg/mL、3mg/mL、16 mg/mL、4 mg/mL。茶皂素具有较好的热稳定性。洗手液试品的感官指标,理化指标及微生物指标均符合或优于相应的国家标准。

Study on Extraction and Antimicrobial Activity of Tea Saponin and Its Application
Abstract:This study optimized the extraction conditions of the defatted tea seed cake, combined with water and alcohol extraction system and the macroporous resin method. Microplate reader was used to measured the effect of Tea Saponin on the growth of several common microbial and determined the minimum inhibitory concentration (MIC); Using turbidity method to detected the thermal stability of tea saponin. The typical recipe of the hand sanitizer was determined by the orthogonal experimental design method, and detected about sensory indicators, physicochemical and microbial indicators. The result shows that: water extraction conditions: extraction temperature 70℃, liquid-solid ratio 8:1, extraction time 4h. The tea saponin having a good antimicrobial effect on seven kinds of experimental bacteria, particularly on the bacteria, the MIC values were: 0.5mg/mL, 1mg/mL, 2mg/mL, 2mg/mL, 3mg/mL, 16 mg / mL, 4 mg / mL. The tea saponin showed strong thermal stability against heat treatment. Sensory、physicochemical and microbiological indicators of hand sanitizer test product are in line with or better than national standards.
