噬菌体抗体库的原理和应用简介

摘要:噬菌体抗体库技术是近年来发展起来的一项技术。它是将体外克隆的抗体基因片段插入噬菌体载体,转染工程细菌进行表达,然后用抗原筛选即可获得特异的单克隆噬菌体抗体。利用这一技术可以得到完全人源性的抗体,在HIV等病毒感染和肿瘤的诊断与治疗方面有其独特的优越性。越来越多的证据表明该方法是一种重要的能进行多种生物学功能分析的技术。本综述简明介绍了噬菌体抗体库的构建过程,并对相关技术细节进行了分析。同时对该方法的当前研究进展进行了介绍,为相关研究提供一定的参考。

Brief introduction of phage antibody library and its application
Abstract:Phage antibody library technology is a recently developed technology. Antibody gene fragment cloning in vitro was inserted into the phage vector, transfection of engineering bacteria for expressing, then with the screening of antigen, specific monoclonal phage antibodies could be obtained. Fully human source antibody could be obtained using this technology, it has unique superiority in the HIV virus infection, tumor diagnosis and treatment. Increasing evidence indicates that this technology is a powerful tool for investigation of various biological functions. This review will discuss the technique details regarding the construction of phage display library and also introduce concisely the application value of this technology, aiming at provide references for related studies.
