蚕蛹油对葛根素口服生物利用度的影响研究

摘要:目的:研究葛根素与蚕蛹油的悬浮液中葛根素在小鼠体内的生物利用度。方法:以葛根素与0.5%的羧甲基纤维素钠的悬浮液为对照,分别小鼠灌胃葛根素羧甲基纤维素钠悬浮液和葛根素蚕蛹油悬浮液(灌胃剂量为葛根素200 mg/kg),采用高效液相色谱法测定小鼠血清中葛根素的含量。结果:药-时曲线数据经DAS软件处理,对照组的tmax、Cmax和AUC0-10h值分别为35.83±5.48 min、0.79±0.18 μg/mL和1.79±0.097 μgoh/mL,而葛根素蚕蛹油悬浮液组的tmax、Cmax和AUC0-10h的值分别为35.71±4.08 min、3.72±0.68 μg/mL和7.00±1.03 μgoh/mL,葛根素蚕蛹油悬浮液的相对生物利用度为391%。结论:与葛根素羧甲基纤维素钠悬浮液相比,葛根素蚕蛹油悬浮液可以显著提高葛根素在小鼠体内的口服吸收。

Study on the effect of silkworm pupal oil on the oral absorption and bioavailability of puerarin
Abstract:The main purpose of this study was to investigate the relative bioavailability of puerarin suspensions in 0.5%CMC and in silkworm pupal oil (equivalent to 200 mg/kg of puerarin) in mice. the puerarin concentrations in blood were determined by HPLC. The pharmacokinetics parameters and the relative bioavailability of the two suspensions were analyzed by the software DAS. Their pharmacokinetics parameters tmax, Cmax, and AUC0-10h were 35.83±5.48 min,0.79±0.18 μg/mL,1.79±0.097 μgoh/mL and 35.71±4.08 min,3.72±0.68 μg/mL,7.00±1.03 μgoh/mL, respectively. The AUC0-10h of the puerarin suspension in silkworm pupal oil after oral administration was markedly higher than that obtained with puerarin suspension in CMC (7.00±1.03 μgoh/mL vs. 1.79±0.097 μgoh/mL, P < 0.01). The difference in AUC is significant. And the relative bioavailability of the puerarin suspension in silkworm pupal oil was 391%. The silkworm pupal oil suspension can effectively improve bioavailability of puerarin through oral administration in mice .
