草鱼、白鲢、鳙鱼和青鱼酸溶性胶原蛋白主要理化性质的比较

摘要:草鱼(Ctenopharyngodon idella),白鲢(Hypophthalmichthys molitrix),鳙鱼(Hypophthalmichthys nobilis)和青鱼(Mylopharyngodon piceus)是国内主要的淡水鱼种。本研究旨在从四种主要淡水鱼的鱼皮、鱼鳞中提取相应的酸溶性胶原蛋白(ASC),并比较他们在结构和热稳定性上的主要差异。研究揭示四种淡水鱼的鱼皮及鱼鳞中提取出的酸溶性蛋白主要为I型胶原蛋白,并含有三条结构不尽相同的α链。经过V8蛋白酶水解后,鳙鱼和白鲢的鱼皮、鱼鳞酸溶性胶原蛋白呈现相似的肽段图谱。同样,草鱼和青鱼的肽段图谱也非常相似,但和前两种鱼的肽段图谱存在差异。通过差示量热扫描法(DSC)测量发现,所提取的酸溶性胶原蛋白的变性温度基本都在34.1-36.4 oC之间。这些结果表明,虽然这四种鱼的鱼皮、鱼鳞酸溶性胶原蛋白在一级结构上存在一定差异性,但他们具有相似的热稳定性,可以同时运用于食品、医药、化妆品等行业。

Extraction and comparison of acid-soluble collagens from the skins and scales of grass carp, silver carp, bighead carp and black carp
Abstract:Grass carp (Ctenopharyngodon idella), silver carp (Hypophthalmichthys molitrix), bighead carp (Hypophthalmichthys nobilis) and black carp (Mylopharyngodon piceus) are the four major freshwater fish species in China. The present study characterized the acid soluble collagens (ASC) from skins and scales of these four species, and compared their similarities and differences in primary structure and thermal stability. All ASC were mainly characterized as type I collagen with three different α chains. After V8 protease digestion, the ASC from both skins and scales of bighead carp and silver carp showed similar peptide hydrolysis patterns, and the ASC from grass carp and black carp were similar to each other but different from the former two species. The values of Tmax of the ASC were measure by DSC and ranged from 34.1 to 36.4 oC. These results suggest that although the ACS from skins and scales of the four fishes are slightly different in primary structure, they show similar thermal stability and all can be used as alternative sources of collagen for food, pharmaceutical and other applications.
