蛋白质磷酸化修饰对卵母细胞成熟的调控作用

摘要:目的:鉴定卵母细胞成熟过程中发生磷酸化修饰改变的蛋白质,并阐述磷酸化修饰的改变介导的蛋白质变化对卵母细胞成熟的调控作用。方法:利用凝胶分析软件比对卵母细胞成熟过程表达差异蛋白质图谱和MII期卵母细胞磷酸化修饰蛋白质图谱数据,找出卵母细胞成熟过程中发生磷酸化修饰改变的蛋白质。并利用生物信息学工具系统注释这些蛋白质的功能,并分析其参与的调控网络。结果:通过分析鉴定到9种蛋白质在卵母细胞成熟过程中发生磷酸化修饰的改变,这些蛋白质大部分具有酶活性并组成复杂调控网络。结论:该研究揭示了卵母细胞成熟过程中磷酸化修饰改变的蛋白质可能参与调控卵母细胞的成熟和早期胚胎发育,并为进一步研究卵母细胞成熟和功能的调控机制奠定了基础。

The Role of Protein Phosphorylation in Oocyte Maturation
Abstract:Objective: To identify quantitatively changed proteins which are modified with phosphorylation during oocyte maturation, and to study the regulatory role of protein phosphorylation changes during oocyte maturation. Method: By using gel electrophoresis image analysis software, phosphorylation changed proteins during oocyte maturation were identified from maps of differentially expressed proteins and phosphorylated proteins in MII oocyte. And bioinformatics tools were used to analyze the function and regulatory network of these proteins.Results: A total of 9 proteins were identified as phosphorylation changed proteins during oocyte maturation, most of them had catalytic activity and evolved in a complex regulatory network. Conclusion: This study reveals that protein phosphorylation change may play a role in the regulatory of oocyte maturation and early embryonic development. It will enable us to further research the mechanism of oocyte maturation and function.
