一种基于二氟硼衍生物的细胞内pH荧光成像探针

摘要:本文设计合成了一种以二氟硼衍生物为母体的新型pH荧光探针DFBP。该探针对pH呈可逆响应,并可实现"裸眼"检测。探针结构为给体(D, donor)-π-受体(A, accepter)模式,其中二氟硼结构缺电子的特点,使其作为电子受体,而供体部分则为具有双供电子基的邻氨基苯硫酚,其结构中含有两个供电子基团-SH和-NH2。DFBP的pKa为5.60,在pH<5.0时呈红色,pH>6.0时呈黄色,荧光强度随着pH的增大而增强,对pH的响应范围在4.0-8.0之间,说明DFBP可用于监测弱酸性条件下的pH变化。此外,DFBP的斯托克斯位移达到了143 nm,可以有效避免检测过程中激发光带来的干扰。将DFBP用于人肝癌细胞内pH原位成像,表明该探针具有良好的膜透性和生物兼容性。

A difluoro boron (BF2) derived fluorescent probe for intracellular pH imaging
Abstract:In this paper, a difluoro boron (BF2) derived fluorescent probe DFBP for cellular pH was designed and synthesized. The probe was reversible and can be used as a naked eye chemosensor. DFBP is characterized of a molecular skeleton of donor-π-accepter, in which the BF2 moiety is electron deficient and the sulfydryl and amino from an o-amino thiophenol moiety are electron rich. This results in an electron push-pull construction of DFBP. The pKa of DFBP is calculated to be 5.60. The fluorescence intensity of DFBP increases with pH augment in the pH range of 4.0~8.0 and it turns red (pH<5.0) and yellow (pH>6.0) at different pH regions, indicating that DFBP is sensitive to minor pH variation under weakly acid conditions. Moreover, the Stokes shift of DFBP is as large as 143 nm, which contributes to the shielding of excitation light during detection. The probe showed good photostability and cell membrane permeability, as well as low cytotoxicity. The probe was successfully applied to the imaging of pH in human HG2 cells with good sensitivity and selectivity.
