TRAPP复合体亚基Trs85与其它TRAPP亚基之间的互作

摘要:早先认为酵母体内的TRAPP(蛋白转运颗粒)复合体存在TRAPPI和TRAPPII两种不同的形态,并将Trs85和Trs33归类为TRAPPI复合体的亚基,但后来的研究发现Trs33属于TRAPPII复合体的亚基。为了澄清Trs85在TRAPP复合体中的归属,本文通过采用酵母双杂交的方法检测Trs85与TRAPP复合体中其它亚基之间可能存在的蛋白互作来进行初步鉴定。实验结果显示Trs85选择性与TRAPPI亚基互作而与TRAPPII亚基不发生互作,说明Trs85不属于TRAPPII亚基而与TRAPPI亚基发生紧密联系。我们的结果与最近所报道的Trs85通过与TRAPPI亚基互作构成新的TRAPPIII复合体后通过小G蛋白Ypt1而不是Ypt31/32调控细胞自噬与囊泡运输的结论一致。

Interactions between Trs85 and other TRAPP complex subunits
Abstract:TRAPP (Transport Protein Particle) complex was reported as a multi-subunit complex existing in two subcomplexes, TRAPPI and TRAPPII. Trs33 and Trs85 were categorized to TRAPPI subunits although Trs33 was later found to be more like a TRAPPII subunit. In order to clarify whether Trs85 belongs to TRAPPI or TRAPPII, we examined the possible interactions between Trs85 and other TRAPP complex subunits by yeast two hybrid assay. Our results showed that Trs85 selectively interacts with TRAPPI subunits, but not with TRAPPII subunits. These results indicate that Trs85 does not belong to TRAPPII complex, but closely associates with TRAPPI complex. Our findings are consistent with the recent reports of Trs85 together with TRAPPI subunits to form new TRAPPIII complex which regulates autophagy and vesicle transport through Ypt1, but not Ypt31/32.
