蹄蝠科3属蝙蝠mtDNA控制区的分子特征与进化

摘要:翼手目蹄蝠科蝙蝠线粒体控制区结构与进化特征少有研究,为了给出蹄蝠控制区详尽的进化特征,本研究首次测序并分析了蹄蝠科3属蝙蝠的控制区全序列。蹄蝠科蝙蝠控制区具有典型的哺乳动物特征,可划分为ETAS、CD和CSB结构域。在ETAS区,检测到ETAS1和ETAS2保守元件,前者变异度(41.9%)低于后者(51.7%);在三叶蹄蝠和双色蹄蝠内检测到长重复元件(分别为26 bp和80 bp),可形成稳定的二级结构。在CD区,共检测到5个保守元件,F、E、D、C和B。在CSB区内,共检测到CSB1、CSB2、CSB3和CSB1-like元件。CSB2变异度最高(44.4%),CSB3最保守。在CSB内还检测到长短不等的串联重复元件(6 bp、8 bp、16 bp或20 bp),蹄蝠科3个属间重复序列的长度、碱基组成和拷贝数均存在差异。

Molecular characteristics and evolution of the mitochondrial control region in three genus of Hipposideridae
Abstract:There have been few studies of the structural and evolutionary characteristics of the mitochondrial control region (CR) in hipposiderids. The CR was firstly sequenced for three genera in Hipposideridae to give a detailed overview of its features. The CR of leaf-nosed bats was divided into three domains (ETAS, CD and CSB) like that of other mammals. In the ETAS domain, ETAS1 and ETAS2 elements were detected, and the variability of ETAS1 (41.9%) was lower than that of ETAS2 (51.7%). Repeated motifs were examined in Aselliscus stoliczkanus (26 bp) and Hipposideros bicolor (80 bp) in ETAS, which can be formed the stable secondary structures. In the CD domain, 5 conserved blocks, F, E, D, C and B, were found. In the CSB domain, CSB1, CSB2, CSB3 and CSB1-like elements were detected in all species. The CSB2 element had the highest variability (44.4%), but the CSB3 was the most conserved, with no variable site. Repeated motifs were present in the CSB in all individuals (6 bp, 8 bp, 16 bp or 20 bp), but the length, base composition and copy number were variable among three genera of Hipposideridae.
