摘要:本文依据Oxytricha nova端粒DNA序列G4T4G4设计了一系列5'末端依次相差一个鸟嘌呤碱基的富G核酸序列d[G4T4G4]、d[G3T4G4]、d[G2T4G4]以及d[T4G4T4G4],定量计算热力学参数ΔH、ΔS和ΔG25,研究末端碱基对这些富G序列与其互补链之间结构竞争的调控。对d[G4T4G4]序列,5'端减掉鸟嘌呤或增加胸腺嘧啶均能导致四链的热稳定性下降,平衡向双链移动。

Regulation of structural competition in Oxytricha nova telomeric DNA
Abstract:In this paper, the role of terminal residues variation in modulating the structural polymorphism of G-quadruplex and the quadruplex-Watson-Crick duplex competition is investigated using four G-rich sequences with two G-tracts involving d[T4G4T4G4], d[G4T4G4], d[G3T4G4] and d[G2T4G4] from Oxytricha nova telomeric DNAs. Effects of terminal variations on the structural competition between the G-quadruplex and the Watson-Crick duplex are investigated quantitatively by thermodynamic parameters (ΔH, ΔS, ΔG25). The relative free energy change shows that decreasing guanines or increasing thymines at 5' end of d[G4T4G4] destabilizes the quadruplex but favors the duplex formation.
