不同蚕品种两种类型乙酰胆碱酯酶基因多态性分析

摘要:研究表明昆虫乙酰胆碱酯酶基因(ace)突变会引起抗药性。为了探析家蚕乙酰胆碱酯酶基因突变与家蚕农药抗药性的关系,本研究测试了毒死蜱与残杀威对24种家蚕毒杀的半效致死浓度(LC50 值),然后克隆、测序了这24种家蚕以及另外7种家蚕两种类型的乙酰胆碱酯酶基因,通过分析,在1型乙酰胆碱酯酶基因(ace1)上发现了63个氨基酸突变,在2型乙酰胆碱酯酶基因(ace)2上发现了14个氨基酸突变,这表明2型乙酰胆碱酯酶比1型乙酰胆碱酯酶基因更为保守。同时分析表明,24种家蚕ace突变与家蚕对毒死蜱和残杀威抗性没有直接关系,这些突变的诱因及功能有待于进一步分析。

Gene polymorphism analysis for two types of acetylcholinesterase of different kinds of silkworm
Abstract:It was reported that some mutations in the acetylcholinesterase(AChE) gene of insect was responsible for pesticides resistance. To examine if ace gene mutation had a role in resistance in Bombyx mori. strains, this study tested the LC50 values of chlorpyrifos and propoxur on 24 strains of B. mori, then we cloned and sequenced the AChE gene from 31 strains. The LC50 values of chlorpyrifos and propoxur on different strains of B. mori with distinguished change, and gene mutations was been found. But these mutations lack of correlation with chlorpyrifos and propoxur resistance, and based on the results of phylogeny analysis, we can conclude that ace2 is more conserved than ace1. The causes of these mutations and the function of the mutated AChE need to be determined for further.
