抑制性寡聚脱氧核苷酸的研究进展

摘要:抑制性寡核苷酸(inhibitory ODN)是一种人工合成的具有特定核苷酸顺序的单链DNA分子,是Toll样受体拮抗剂,能抑制由内源性或外源性TLR配体引起的免疫细胞的激活反应。抑制性ODN的免疫抑制活性与其自身的结构特点有关,其作用机理与DNA分子骨架的硫代修饰、ODN的长度和核苷酸序列的特异性等相关。抑制性ODN作为一种靶点明确的信号阻断剂,已经成功成功应用于自身免疫性疾病和脓毒血症、多发性硬化及风湿性关节炎等与TLR相关的疾病模型的研究中。因而研发新型的寡核苷酸类药物,具有很强的现实意义与经济意义。

Advance of inhibitory ODN
Abstract:The inhibitory oligonucleotides (inhibitory ODN) is a synthetic single-stranded DNA molecule, , as Toll-like receptor antagonists which can inhibit the reaction caused by endogenous or exogenous TLR ligand. Different inhibitory ODN function by each own structural features,such as DNA molecule skeleton phosphorothioate, and the length and nucleotide sequence of ODN specificity related. Inhibitory ODN has been successfully applied to TLR related diseases model study , such as autoimmune diseases and sepsis, multiple sclerosis and rheumatoid arthritis. Thus a new type of oligonucleotide drugs would be hopeful therapy treatment
