甲型冠状病毒属三个病毒株对不同宿主来源细胞的感染性比较

摘要:冠状病毒的重组和变异频率很高,本文比较了甲型冠状病毒属G1亚群的代表毒株犬冠状病毒(CCoV1-71)、猫冠状病毒(FCoV 79-1683)、猪传染性胃肠炎病毒(TGEV pudue)在不同宿主来源细胞上的感染特性。病毒交叉感染犬、猫和猪来源的细胞系后,比较病毒在不同细胞中的增殖,细胞病变及噬斑形态。结果发现,三种病毒均能在其相应的自然宿主动物来源的细胞上增殖良好,并具有典型的细胞病变,形成较大的噬斑。而三种病毒在非宿主来源的细胞上则表现出差异性:三种病毒均能在犬源的A72细胞上增殖良好, CCoV在猫源的FCWF细胞中增殖强于FCoV,而CCoV和FCoV在猪源的ST细胞中增殖水平明显受到了限制。本文的研究结果表明,甲型冠状病毒G1亚群间有较强的宿主适应能力,因而出现共感染和重组几率高。

The replication of three members of Alphacoronavirus 1 in cultural cells originated from different host animals
Abstract:The replication characters of alphacoronaviruses, including CCoV, FCoV and TGEV, were studied in different original cell lines to understand the abilities of those viruses to cross host barrier. A72, FCWF and ST cells were infected by CCoV, FCoV and TGEV respectively. The samples of cell cultures were collected at different post-infection pionts. Plaque-assay was carried out to detect the virus titers. The configuration of plaques and the CPE induced by three viruses were observed. Results: The results showed that those viruses replicated effectively in their natural hosts with typical CPE, bigger and good shape plaques, and highest titers of these viruses appeared at post-infection 12 hours. But in no-natural hosts, those three virues showed different characters. The plaques showed that CCoV and FCoV infected A72 cell more effectively than TGEV. FCoV have a higher virulence to FCWF than CCoV, and the titer of TGEV was the lowest. Distinct discriminations were found in ST cell. FCoV and CCoV could not replicate well in ST cells with lower titers and unformed plaque, indicating that those two viruses could not adapted to ST cell. It proved that the coronaviruses intragenus could easily break the species-barrier of each other, but difficult intergenus.
