拟南芥Villins基因在花器官中的表达分析

摘要:Villin(VLN)是一类肌动蛋白结合蛋白,参与调节微丝骨架的动态变化。采用启动子(包含第一个内含子)融合GUS基因的方法, 研究了VLN1-5在拟南芥花器官中的表达。结果表明:1)在花的不同组织器官中,有不同的VLN基因表达,说明VLN基因参与花器官的发育。2)不同的VLN基因表达具有较高的特异性。VLN1在除花粉粒之外的花器官内都表达;VLN5在除花萼、胚珠外的花器官内都表达;VLN2只在花萼中表达。VLN1、VLN3、VLN4均在花柱中尤其是柱头中表达,但只有VLN1在柱头的乳突细胞中表达;VLN5在花粉粒和柱头的乳突细胞中高量表达。推测VLN1和VLN5可能参与拟南芥授粉过程中柱头的乳突细胞和花粉间的相互作用。研究的结果有助于进一步研究VLN调控微丝骨架动态在植物授粉过程中的功能。

The differential expression patterns of Arabidopsis villin genes in floral organs
Abstract:Villins (VLNs) are actin binding proteins that regulate actin dynamics in plant cells. There are five VLN genes in Arabidopsis. We expressed respectively the promoters (including the first intron sequences) of VLN1-5 and GUS in Arabidopsis via Agrobacterium-mediated transformation. The expression patterns of VLN1-5 are different, according to the GUS histochemical staining assay. All VLN genes are expressed in floral organs, which indicate that VLNs maybe involve in flower development. VLN2 is expressed only in the calyces. VLN1, VLN3, VLN4, and VLN5 are expressed in the style and stigma, however only VLN1 and VLN5 are expressed in the papillate cells. Moreover, VLN5 is also expressed in the pollen grains. These results suggest that VLN1 and VLN5, especially the latter, maybe function in the interaction between pollen grain and papillate cell during the pollination of Arabidopsis though the actin dynamics.
