猪S100A8、S100A9基因组织表达分布及真核表达载体构建

摘要:S100A8、S100A9隶属于S100钙结合蛋白家族,主要在粒细胞、巨噬细胞、单核细胞等髓源性免疫细胞以及炎症反应相关的内皮细胞中表达,正常情况下S100A8、S100A9基因低表达,而当病原感染时S100A8、S100A9基因急剧上调表达并且以复合体形式分泌到胞外,通过与TLR4结合诱导机体炎症反应,在机体抵抗病原感染过程中发挥重要的作用。猪S100A8、S100A9基因紧密连锁,在副猪嗜血杆菌感染及LPS、Poly(I:C)刺激时猪S100A8、S100A9基因显著上调表达,这表明S100A8、S100A9基因在猪感染中发挥着重要作用。本研究检测了猪S100A8、S100A9基因组织表达分布,并构建了猪S100A8、S100A9基因真核表达载体。这些研究为揭示猪S100A8、S100A9基因生物学功能奠定了基础。

Investigation of tissue expression pattern and eukaryotic expression vector construction of porcine S100A8 and S100A9 genes
Abstract:The S100A8 and S100A9 genes are belong to S100 calcium binding protein family. They were mainly expressed at marrow origin cells such as granulocytes, macrophages and monocytes. Also, they were expressed at the endotheliocytes involved into inflammatory response. The S100A8 and S100A9 genes were up-regulated when the body was infected. They two proteins can form a complex and bind to TLR4 to induce the inflammatory response when they were secreted out of the cells. The two proteins played very important roles in the resistance against infection. The porcine S100A8 and S100A9 were closely linked on the genome and were up-regulated after HPS infection or stimulated with LPS/Poly (I:C). S100A8 and S100A9 may also play important roles in disease resistant in pig. In this study, we detected the tissue distributions of S100A8 and S100A9 genes in pig. Also, the overexpression vectors of porcine S100A8 and S100A9 genes were constructed. Our study offered new information and vectors for further studying the functions of these two genes.
