UVB诱导人皮肤成纤维细胞凋亡与胱天蛋白酶-8关系的研究

摘要:目的: 在UVB诱导皮肤成纤维细胞急性损伤过程中,探讨细胞凋亡及其与胱天蛋白酶-8(caspase-8)的关系。方法:人皮肤成纤维细胞经100、200、300和400mJ/cm2 UVB照射后48小时,用MTT法检测细胞活性变化,用Hoechst 33258染色法检测细胞凋亡,并用抑制剂Z-IETD-FMK抑制 caspase-8活性后,检测凋亡细胞数量。结果:(1)100、200、300和400mJ/cm2 UVB能够明显抑制成纤维细胞活性,引起细胞凋亡,随着照射UVB剂量的增加,凋亡细胞逐渐增多;(2)抑制剂Z- IETD -FMK能够抑制UVB照射引起的细胞凋亡。结论:UVB照射能够诱导皮肤成纤维细胞出现凋亡,在这一过程中caspase-8发挥着重要作用。?????

Study of the relationship between cell apoptosis and caspase-8 in ultraviolet B-damaged human dermal fibroblasts
Abstract:Objective: To explore whether cell apoptosis appeared and the relationship between cell apoptosis and caspase-8 in UVB-induced damage of human skin fibroblasts. Methods: Primary human skin fibroblasts were cultured in vitro and subcultured, 3 to 6 passages of cells were used in experiments. Cell viability affected by 100, 200, 300 and 400mJ/cm2 UVB was measured by the 3-(4,5)- dimethylthiahiazo(-z-y1)-3,5-diphenytetrazoliumromide (MTT) assay at 48 hours after irradiation. Through Hoechst staining, the morphological changes of nuclear in cells treated by UVB without/with caspase-8 inhibitor (Z-IETD-FMK) were observed and the numbers of apoptotic cells were counted. Results: When cells were treated with 100, 200, 300 and 400mJ/cm2 UVB, cell viability decreased with the increasing of UVB dose. UVB could induce cell apoptosis, and when the activity of caspase-8 was inhibited by Z-IETD-FMK, UVB-related apoptotic cell numbers decreased. Conclusion: In the process of acute photo-damage, UVB could trigger apoptosis of human dermal fibroblasts, and the role of caspase-8 was important.)
