CYP19A基因对牙鲆(Paralichthys olivaceus)卵巢发育周期的表观遗传调控机制

摘要:此研究以成年雌性牙鲆(Paralichthys olivaceus)为研究对象,采用荧光实时定量方法检测不同卵巢发育期的性腺P450aromA (CYP19A) 酶基因表达,运用重亚硫酸氢盐修饰后测序法测定其启动子区CpG富集区每个CpG位点甲基化模式。结果发现,在整个卵巢发育期都有CYP19A 基因表达,在Ⅳ期达到最高值,然后下降至卵巢发育初期水平;而启动子CpG富集区整体甲基化水平却与之成相反趋势。这暗示P450aromA酶是催化雌激素合成的关键酶,促进卵母细胞成熟;CYP19A 基因表达可能受启动子区CpG甲基化修饰影响。这些结果初步解释了CYP19A 基因的生理功能及其表达调控的分子机制。

Epigenetic regulation mechanism of CYP19A gene on ovary reproductive cycle of Japanese flounder (Paralichthys olivaceus)
Abstract:The adult female Japanese flounder was obtained in this study. Quantitative PCR was used for detecting the gene expression of CYP19A gene during the reproductive cycle, and bisulphate PCR was performed for described the DNA methylation pattern of CpG rich region in CYP19A gene promoter domain. The result founded the CYP19A gene was expressed in the period of oocyte maturation. In stage Ⅳ was reached the highest level, then decreased in stage Ⅴ, While, the opposite tendency was showing in the DNA methylation level of promoter. This implied that P450aromA enzyme play important role in catalyzing synthesis of estrogen and promotes oocyte maturation; DNA methylation modification of CpG sites in promoter may affect the gene expression. These findings preliminary explain the function of CYP19A gene and the molecular mechanism of expression regulation.
