绿色荧光蛋白转基因昆明白小鼠的建立

摘要:目的 建立表达绿色荧光蛋白(GFP)的转基因昆明白小鼠,为相关实验提供带报告基因的远交系动物模型。方法 用携带有绿色荧光蛋白报告基因的慢病毒载体感染昆明小鼠胚胎干细胞系KES,得到稳定高效表达绿色荧光蛋白的胚胎干细胞KES-GFP。通过将KES-GFP与129品系小鼠的胚胎进行聚合发育形成嵌合体囊胚,移植到假孕小鼠子宫获得表达绿色荧光蛋白的嵌合体小鼠,嵌合体小鼠与昆明白小鼠交配获得转基因小鼠。结果 成功获得了表达GFP的转基因昆明白小鼠。结论 基于胚胎干细胞成功建立了绿色荧光蛋白转基因昆明白小鼠。

Establishment of Transgenic Kunming White Mice Expressing Green Fluorescence Protein
Abstract:AIM To establish transgenic Kunming white mice expressing green fluorescence protein (GFP), providing outbred animal models carrying reporter gene for related studies. METHODS Infected Kunming mouse embryonic stem cell line KES using a lentivirus carrying GFP as a reporter gene, and generated an ES cell line KES-GFP expressing GFP efficiently and stably. These ES cells were introduced into host embryos from 129 stain mouse through aggregation to form chimeric blastocysts, then transferred it into uterus of pseudopregnant mice to obtain chimeric mice expressing GFP. These chimeric mice were mated with Kunming mice to obtain transgenic mice. RESULTS We obtained transgenic Kunming white mice expressing GFP successfully. CONCLUSION Transgenic Kunming white mice expressing GFP can be successfully established based on embryonic stem cells.
