A Sensitive Electrochemical Impedimetric Morphine Immunosensor

Abstract:In this paper, an immunosensor based on self-assembly modified (SAM) Au electrode for the specific detection of morphine (MO) is reported. By the strategy of 3-mercaptopropionic acid SAM, activated by 1-ethyl-3-(3-dimethylaminoprophyl) carbodiimide (EDC) / n-hydroxysulfsuccinimide (NHS) and the immobilization of abundant morphine antibody, the proposed immunosensor provides the sensing signal based on its specific covalent binding with mophine. The technique of electrochemical impedance spectroscopy (EIS) is used as the read-out signal. The value of electron transfer resistance (Ret) responds upon the concentration of mophine with a detection limit of 0.266 pg/L. The immunosensor can be regenerated for at least 2 times by the elution with NaOH-H3PO4 (0.1 mol/L, pH=13.0). When it was used to detect the MO in spiked serum samples, there were satisfactory recoveries between 84.1% and 110.9%. In conclusion, the proposed sensor is proven to be more advantageous compared to those once reported methods in terms of the sensitivity and simplicity.

摘要:本文报道一种基于自组装电极的吗啡免疫传感器,基于3-巯基丙酸自组装、EDC/NHS活化并固定足够量吗啡抗体,所研制的免疫传感器基于对吗啡的特异性共价键合提供了传感信号。以电化学阻抗作为信号读出技术,其电子转移阻抗(Ret)值对吗啡浓度有灵敏响应,检出限为0.266 pg/L。该传感器用NaOH-H3PO4 (0.1 mol/L,pH=13.0)可再生使用两次以上,将此传感器应用于加标血清样品的测定,所得回收率在84.1%至110.9%间,令人满意。该传感器较文献报道方法有更高的度,使用更为简单便捷。
