靶向干预自噬基因Beclin-1对胶质瘤细胞C6放射敏感性影响的研究

摘要:目的 研究自噬基因Beclin-1对胶质瘤细胞C6放射敏感性的影响。方法 构建靶向Beclin-1 shRNA慢病毒载体,感染胶质瘤细胞C6,RT-PCR、Western blot检测 Beclin-1的下调效率,克隆形成实验检测靶向干预Beclin-1对胶质瘤细胞C6放射敏感性的影响。结果 Beclin-1 shRNA慢病毒感染C6细胞后,Beclin-1基因及蛋白表达水平较对照组及空载体组显著下降(P﹤0.05),克隆形成实验中对照组及空载体组C6细胞存活分数(SF2)为0.93及0.91,明显高于Beclin-1 shRNA慢病毒感染组(SF2=0.58)。结论 靶向干预自噬基因Beclin-1可显著提高胶质瘤细胞C6的放射敏感性。

The effects of target interference autophagy-associated gene Beclin-1 to the radiation sensitivity of C6 glioma cells
Abstract:Objective To investigate the effects of target interference autophagy-associated gene Beclin-1 to the radiation sensitivity of C6 glioma cells. Methods Construct Beclin-1 shRNA vector and investigate the interference efficiency by RT-PCR and Western blot. Furthermore, detect the effects of target interference Beclin-1 to the radiation sensitivity in C6 glioma cells by colony forming assay. Results Beclin-1 shRNA vector could significantly repress the level of Beclin-1 mRNA (P﹤0.05) and protein (P﹤0.05) in C6 glioma cells. The SF2 of C6 glioma cells in control group and negative control group (0.93 and 0.91) were much higer than in Beclin-1 shRNA group (0.58). Conclusion Target interference autophagy-associated gene Beclin-1 can radiosensitize C6 glioma cells.
