冬眠对达乌尔黄鼠骨骼肌超微结构的影响

摘要:目的:研究冬眠对达乌尔黄鼠(Spermophilus dauricus)比目鱼肌和趾长伸肌超微结构的影响。方法:黄鼠分为活跃期(GA)、冬眠期(GH)和出眠时(即出眠1~2天的黄鼠)(GAH)共三组,透射电子显微镜法观察骨骼肌纤维的超微结构。结果:1、与活跃期相比,黄鼠比目鱼肌在冬眠中和出眠时梭内肌纤维超微结构无明显改变,而趾长伸肌在此时期仅梭内肌纤维外囊膜有一定的溶解破坏,总体结构仍保持完整;2、与活跃期相比,冬眠中黄鼠比目鱼肌和趾长伸肌的梭外肌纤维出现了少量的粗细肌丝排列紊乱,肌原纤维丢失、溶解现象,肌节变短,肌纤维中糖原颗粒增加,出现线粒体聚集。结论:黄鼠骨骼肌超微结构在冬眠中出现轻微的退行性改变。

Effects of hibernation on ultrastructure of skeletal muscle in ground squirrels
Abstract:Aim: To study the effects of hibernation on skeletal muscle fiber ultrastructure in a ground squirrel model. Methods: 24 animals were allocated to three groups (N=8 each): non-hibernating control (Pre-H), animals hibernated for 112±14 days (H), and animals 2 days after arousal (Post-H). Ultrastructural changes of soleus (SOL) and extensor digitorum longus (EDL) muscles were investigated by transmission electron microscopy. Results: (1) No significant ultrastructural changes occurred in SOL muscle spindles with hibernation. In EDL muscles, a vacuolar phenomenon was observed, plus decomposition in the outermost layer of some muscle spindle capsules. Intrafusal fibers and cross-sectional stripes disappeared or became hard to distinguish in H and Post-H samples. (2) Sarcomere length decreased and numbers of myofiber glycogen increased from compared to Pre-H in the H group. Assembled mitochondria under the sarcolemma were found only in SOL. Conclusions: Hibernating ground squirrel skeletal muscle fibers showed a little degressive changes.
