鹦鹉热衣原体PmpD N端重组蛋白诱导SPF鸡的免疫保护效果评价

摘要:鹦鹉热衣原体多型外膜蛋白(Polymorphic membrance proteins,POMPs)是位于衣原体细胞膜表面,且功能未知的蛋白家族,其中PmpD-N端蛋白属于POMPs一个组成部分,编码蛋白分子量约为64KD。为了探讨禽鹦鹉热衣原体PmpD-N端蛋白的生物学功能,我们克隆、表达、纯化了重组的禽鹦鹉热衣原体PmpD-N端蛋白,与弗式佐剂配合制备了PmpD-N端亚单位疫苗。以高剂量组(100μg/只)和低剂量组(50μg/只)免疫21日龄SPF鸡,同时以商品化的鸡衣原体亚单位基因工程疫苗(50μg/只)为阳性对照,以疫苗佐剂空白对照组。间隔14天进行2次免疫。分别免疫后的7d和14d采集血并分离出血清,以间接酶联免疫吸附试验(ELISA)检测特异性抗体水平;同时,采集外周血液并分离淋巴细胞,用T淋巴细胞增殖(MTT法)试验评价淋巴细胞增值水平。第2次免疫14天后以鹦鹉热衣原体CS1株进行攻毒,攻毒后第7d采咽棉拭子,扑杀所有SPF鸡采集肺脏和脾脏组织器官,免疫组化的方法检测衣原体在靶器官的分布和衣原体的排菌情况。结果显示PmpD-N端蛋白以100μg/只可诱发较高的体液免疫水平和细胞免疫水平,且免疫保护效果与鸡衣原体亚单位基因工程疫苗差异不显著,证实重组PmpD-N端蛋白是一个潜在的免疫保护性抗原成分。

Evaluation of immune response elicited by the recombinant PmpD N terminal proteins of Chlamydia psittaci in SPF Chickens
Abstract:Polymorphic membrance proteins(Pmps) are located on the surface of Chlamydia psittaci and the potential functions are unclear so far. Moreover, PmpD N terminal is 64kD and belongs to the Pmp families.In order to evaluate PmpD N terminal, we cloned, expressed and purified the target protein. Subsequently, the recombinant protein vaccines were manufactured with the Frenz adjuvant.Furthermore, 21-day-old SPF chickens were administered with 100μg and 50μg of the recombinant proteins, respectively.After two weeks, the chickens were given the boosting immunization.The birds inoculated with 50μg of the recombinant MOMP were used as the positive group while the chickens with the adjuvant were regarded as the negative control. The blood samples were collected at days 7 post first immunization and the sera were also sampled at days 14 after second inoculation.The antibody levels against PmpD N terminal were detected by ELISA assay while the lymphocytes proliferation was determined by MTT approach.Finally, all birds were challenged intraperiteally with vivulant strain CB8 of C.psittaci and the pathogen excretions were detected using the commercial kits.Mean while, the lungs and spleens were collected and detected the chlamydial loads using the immunochemistry approach.Taken together, 100μg of the recombinant PmpD N proteins could induce a better humoral response and cellular response and no significant difference was found in comparison with the commercial vaccine, suggestion of the potential vaccine candidate against C.psittaci.
