罗布麻多糖对胰岛beta细胞增殖的影响

摘要:多糖是药用植物中主要的活性成分,具有抗肿瘤、降血糖和降血脂等多种药效,罗布麻(Apocynum venetum)是我国分布较广的药食同源植物,目前对于罗布麻多糖的研究甚少。本文将罗布麻叶经过水煮、醇沉、脱蛋白制得罗布麻叶总糖(ALP),经由多步离子交换层析制备出一系列多糖级分,包括1种中性糖级分(ALPN)和3种酸性糖级分(ALPA,ALPA-1,ALPA-2)。MTT结果表明,ALPA-2具有显著地特异性促进胰岛beta细胞增殖的活性,是罗布麻叶多糖中的有效活性级分。结构分析显示ALPA-2为富含聚半乳糖醛酸(HG)结构域的果胶多糖。初步的活性机制研究表明,ALPA-2促进细胞增殖与其增强线粒体功能相关,而与细胞周期无关。此研究结果为深入探讨罗布麻多糖的构效关系提供依据,为罗布麻的应用奠定基础。

Effects of polysaccharides from Apocynum venetum L.on islet beta cell proliferation
Abstract:Polysaccharide is the main active component in the medicinal plants, it has many pharmacological activities, including anti-tumor, anti-diabetic and anti-hyperlipidemia. Apocynum (Apocynum venetum) is a Medicinal and Edible Plant widely distributed in our country. At present, Apocynum polysaccharides are little studied. In this paper, Apocynum leaves total sugar (ALP) was obtained from Apocynum leaves after being boiled, alcohol precipitated and deproteinated. Then, ALP was fractionated into series of polysaccharide fractions via multi-steps of ion exchange chromatography, including one neutral fraction (ALPN), and three acidic fractions (ALPA, ALPA-1 and ALPA-2). MTT assay showed that ALPA-2 specifically promoted beta cell proliferation, suggesting ALPA-2 was the active fraction in Apocynum leaves polysaccharides. Structural features analysis elucidated that ALPA-2 was the pectic polysaccharide with polygalacturonic acid (HG) rich domain. The preliminary mechanism studies indicated that the activity of ALPA-2 was related to its enhancement of mitochondrial function, but not cell cycle. Results of this study would provide a basis to research structure-activity relationship of Apocynum polysaccharide and apply the Apocynum.
