二至丸对CCl4致大鼠急性肝损伤血浆代谢组学研究

摘要:目的 研究CCl4致大鼠急性肝损伤模型大鼠血浆内源性代谢物变化特点和二至丸对异常代谢的调节作用,寻找二至丸治疗急性肝损伤可能的代谢生物标志物,以探索二至丸治疗肝损伤的作用机制。方法 运用快速分离液相色谱-串联四极杆飞行时间质谱(RRLC-Q-TOF/MS)技术,对大鼠血浆中内源性代谢物进行测定。结果 定性鉴定出9种潜在代谢生物标志物,通过独特通路分析(IPA),找到2条肝损伤代谢通路。结论 二至丸可以使造模后大鼠的体内代谢物含量有不同程度的恢复,提示其治疗作用可能与2条肝损伤代谢通路的调节有关。独特通路分析能有效的识别药物靶标,这有助于进一步了解药物作用机制,提高研究效率。

Metabolomics study of Erzhi Pills on acute hepatic injury induced by CCl4 in rats
Abstract:AIM Plasma metabonomics study of Erzhi Pills on acute hepatic injury induced by CCl4 in Rats. To observe plasma metabolic changes of acute hepatic injury induced by CCl4 in rat and the regulating action of Erzhi Pills (EZW) on abnormal metabolism, and to find the potential biomarkers. To explore mechanisms of Erzhi Pills in treatment of acute hepatic injury induced by CCl4. METHODS Endogenous metabolites in plasma were determined with RRLC-Q-TOF/MS. RESULTS Nine potential metabolic biomarkers were identified qualitatively. Two metabolic pathways were identi?ed using ingenuity pathway analysis (IPA). CONCLUSION Changed metabolites can be certainly recovered by Erzhi Pills, and the treatment effect of Erzhi Pills can be connected with the regulation of 2 related metabolic pathways. Unique pathway analysis can be effective to drug-target identification, which helps to understand further mechanism of drug action, and improve research efficiency.
