恒强旋转磁场对骨折愈合过程中BMP-2和PDGFs表达的影响

摘要:目的:观察恒强旋转磁场(RCSMF)促进骨折愈合过程中,骨形态蛋白(BMP)和血小板源性生长因子(PDGFs)在骨折部位表达的变化,探索RCSMF促进骨折愈合的可能机制。方法:80只SD大鼠建立股骨闭合性骨折模型,随机分为磁场干预组(M)和对照组(C),C组无处理,M组置于0.4T RCSMF中,磁体旋转频率6HZ,30分钟/天,每周6次。按骨折后观察时点不同两组大鼠分别又分为2周,3周,4周和8周8个亚组,对应时点处死各组动物,常规组织学处理,骨折部位纵行切片,免疫组化染色测定BMP-2,PDGF-A和PDGF-B在骨折部位阳性表达,并计算阳性区域的累计光密度(IOD)。双因素方差分析比较时间、RCSMF干预的主效应及交互效应,显著性水平为p<0.05。结果:不同时间BMP-2,PDGF-A和PDGF-B阳性表达IOD均出现显著性的变化。RCSMF与时间对BMP-2阳性表达的IOD有交互效应。RCSMF干预可以显著的提高PDGF-B阳性表达的IOD。结论:RCSMF可以通过调节骨折部位BMP-2和PDGF-B的表达,促进骨折部位的修复。

The Effect of Rotation Constancy Static Magnetic Field on BMP-2 and PDGFs During Fracture Healing
Abstract:Objective: Rotation constancy static magnetic field(RCSMF) has recently been demonstrated to promote fracture healing. The aim of this study was to determine the expression of bone morphogenetic protein-2(BMP-2) and Platelet-derived growth factors (PDGFs) in RCSMF promotion of fracture healing, and thereby elucidate the possible roles of the BMP-2 and PDGFs in fracture healing. Methods: 80 Rats with femur closed fracture were randomly divided into magnenic treatment group(M) and control group(C). the M group was given RCSMF treatment using 0.4 T 6HZ magnetic field 30 minutes per day, 6 times per week. Femurs and calluses were subjected to BMP-2, PDGF-A and PDGF-B immunohistochemistry assay 2, 3, 4, and 8 weeks after treatment. The integrated optical density(IOD) of positive staining were caculated. Two-factor variance analysis was used to compare the main effect of time , treatment and the interaction of time×treatment. Significance level was p<0.05. Results: The IODs of immunoreactivity of BMP-2, PDGF-A and PDGF-B were significant among different time group. Time×treatment had signicant interaction effect on BMP-2. RCSMF increased the positive staining IOD of PDGF-B significantly. Conclusion:The findings suggest that BMP-2 and PDGF-B play an important role in RCSMF-activated callus cell proliferation and bone remodeling of fracture healing.
