原位杂交技术检测miRNA-34c在小鼠睾丸和精原干细胞的表达

摘要:目的:研究miR-34c在小鼠睾丸组织和小鼠精原干细胞中的表达规律。方法:应用FISH (Fluorescence in situ hybridization)技术完成在睾丸组织切片和精原干细胞的miR-34c示踪。结果:实验结果表明miR-34c在2月龄小鼠睾丸中表达量较高;在转染入miR-34c 模拟物组miR-34c 表达量较其他组高。结论:使用FISH方法探索miRNA 表达规律较稳定,且重复率高。

miRNA-34c expression in mouse testis and spermatogonial stem cells(SSCs) identification by FISH
Abstract:Objective: To explore the expression of miR-34c in mouse testis tissues and mouse spermatogonial stem cells. Methods: To illustrate the miR-34c expression, microRNA FISH (Fluorescence in situ hybridization) was used. Results: miR-34c expressed higher in 2m mouse testis tissue slices and the group of miR-34c mimics transfected SSCs. Conclusion: To measure miRNA expression, FISH is a stable and repeatable method.
