自噬与Dami细胞增殖分化关系的初步研究

摘要:目的 探讨自噬与Dami细胞的增殖分化的关系。方法 利用PMA刺激Dami细胞建立Dami细胞增殖分化体系,通过流式细胞技术检测Dami细胞多倍体形成和细胞表面CD41表达。利用Western blot检测Dami细胞增殖分化过程中自噬相关蛋白LC3表达,同时观察了自噬抑制剂Bafilomycin A1对Dami细胞增殖的影响。结果 PMA刺激Dami细胞后,细胞核增大并出现多倍体,CD41表达量增加。并且在PMA刺激Dami细胞增殖分化过程中,发现二型自噬相关蛋白LC3的表达量增加。加入自噬的抑制剂Bafilomycin A1阻断自噬溶酶体形成后,可抑制由PMA诱导的Dami细胞增殖,同时伴有细胞凋亡的发生。结论 Dami细胞的增殖分化过程中伴随自噬的发生。

Primary Studies on the Correlations of Autophagy to the Proliferation and Differentiation of Dami Cells
Abstract:Objectives:To study the roles of autophagy in the differentiation and proliferation of Dami cells. Methods and results: The polyploidy and CD41 expression of Dami cells stimulated by PMA were measured by flow cytometry to determine their proliferation and differentiation, respectively, and the autophagy-related proteins LC3 were detected by Western blot. The effect of Bafilomycin A1, an agent to block autophagosome formation, on Dami proliferation and differentiation was also observed. Results: we showed that PMA induces the polyploidy formation and increases CD41 expression of Dami cells accompanied by an enhanced LC3-II expression. Consistently, Bafilomycin A1 inhibited the proliferation of Dami cells induced by PMA and induced apoptosis as well. Conclusions: Our data showed that autophagy is involved in the proliferation and differentiation of Dami cells.
