iTRAQ技术在植物中的应用

摘要:近些年来,随着生物科学技术的不断进步,蛋白质组学研究已经成为生命科学研究的热点,其相关研究技术也如雨后春笋一般涌现,同位素相对标记和绝对定量 (iTRAQ)技术自从2004年被美国应用生物系统公司开发以来,依仗着自身独特的高敏感性和高准确性的串联质谱及多维液相色谱联用,已经逐渐成为蛋白质组学定量分析的主流技术。iTRAQ技术可对复杂样本、细胞器、细胞裂解液等样本进行相对和绝对定量研究,具有较好的定量效果及较高的重复性。由于其能够同时对最多8种样品进行标记分析,使得其在生命科学的各个领域都取得了广泛的应用。在植物的蛋白组学研究中也取得了很大进展。本文对iTRAQ技术的原理、优缺点、实验流程以及在植物中的应用进展进行了综述。

Application of iTRAQ technology in plants
Abstract:In recent years, with the radiply development of biological science and technology, proteomic research has become a hotspot in life science research, and its corresponding methodological research has sprung up like mushrooms, in 2004, A new technique was developed by Applied Biosystems : relative and absolute quantification(iTRAQ) technology, it has high sensitivity and accuracy, particularly the new combination of multi-dimensional liquid chromatography and mass spectrometry (MS) based technologies with protein fragmentation methods, it has gradually become the mainstream of quantitative analysis of proteomics technology. The iTRAQ technology has a good quantitative effect and a high repeatability result in complex samples, organelles, and whole cell lysates. It can simultaneously analysis up to eight samples, hence iTRAQ has been widely used in areas of life science. It also has made great progress in the study of plant proteomics. This paper is focused on the principle, experimental procedure and application in plants of iTRAQ.
