活化-冻干壳聚糖微球固定脂肪酶的研究

摘要:本文采用真空冷冻干燥技术制备了具有空间网状结构的壳聚糖微球,考察了冻干对载体形貌及活性位点的影响;以活化-冻干处理后的壳聚糖微球为载体固定假丝酵母脂肪酶(Lipase EC3.1.1.3),对固定化效果进行了研究,并与直接固定和活化-固定的固定化酶进行了对比,考察了活化过程对固定化酶性能的影响。结果表明,采用活化-冻干-固定法制备的固定化酶性能优良,其固定脂肪酶的水解活力可达到57.2U/g。

The study of immobilization of lipase in activated-freeze-drying chitosan microsphere
Abstract:In this article,vacuum freeze-drying technology was used to prepare the chitosan microspheres, which have three-dimensional spatial network structure. The effects of vacuum freeze-drying on the morphology of microspheres and active sites were investigated. Candida lipase(Lipase EC3.1.1.3) were immobilized on the activate-lyophilized chitosan microspheres. The immobilization performance of these microspheres was studied and compared with direct-immobilization and activate-immobilization. The influence of activate process on the performance of immobilized lipase was also illustrated. The experimental results indicated that using activate-lyophilized chitosan microspheres as carrier to immobilize lipase is an optimal choice, since the activity of immobilized lipase was over 57.2U/mg.
