白藜芦醇通过ERK1/2-CREB信号通路抑制血管紧张素Ⅱ诱导的心肌肥大

摘要:目的:研究白藜芦醇(resveratrol, Resv)对血管紧张素II(Angiotensin II,AngII)诱导H9C2细胞肥大的抑制作用及ERK1/2-CREB信号通路的影响,并探讨其机制。方法:通过建立Ang II 诱导H9C2细胞肥大模型,采用Image Pro Plus 6.1软件测量细胞表面积大小;BCA法测定细胞总蛋白含量,Western-blot方法检测H9C2细胞p-ERK1/2、ERK1/2、p-CREB和CREB蛋白的表达。结果:与对照组相比,AngII(0. 1μM)孵育后,细胞表面积增加,细胞蛋白含量增加了46 %(P<0.01)。Resv (0.01μM +AngII组,0.1μM +AngII组, 1μM +AngII组)能浓度依赖地减少蛋白含量。Ang II 诱导H9C2细胞p-ERK1/2和p-CREB蛋白的表达升高。不同剂量Resv对AngII 诱导H9C2细胞p-ERK1/2和p-CREB蛋白的表达均有抑制作用,并且呈浓度依赖。结论:Resv抑制AngII介导的H9C2细胞肥厚,其机制可能是通过抑制细胞表面积增大、细胞蛋白含量增多和ERK1/2-CREB信号通路实现的。

Resveratrol inhibits angiotensin Ⅱ-induced cardiac hypertrophy via the ERK1/2-CREB signaling pathway
Abstract:Objective: To investigate the inhibitory effects of resveratrol on Ang II-induced cardiac hypertrophy in H9C2 cell, which is helpful to elucidate the ERK1/2-CREB signaling pathway mechanism of cardiac hypertrophy. Methods: Ang II was administered to establish the cardiacmyocytes hypertrophy model of H9C2 cells. The Cell surface area of H9C2 cells was measured by Image Pro Plus 6.1 software. The total protein content of H9C2 cells was detected by BCA method. The protein expressionof ERK1/2, p-ERK1/2, CREB and p-CREB were detected by western blot. Results: compared with the control group, cell surface area was increased significantly in Ang II group (P<0.01). Compared with control group, the total protein content of cells was increased significantly in Ang II group (P<0.01). Resv (0.01μM +Ang II group,0.1μM +Ang II group, 1μM +Ang II group) of total protein content decreased in a concentration-dependent manner. Ang II stimulated phosphorylation of ERK1/2 and CREB. Resv (0.01μM +Ang II group,0.1μM +Ang II group, 1μM +Ang II group) could reduce the expression level of p-ERK1/2 and p-CREB in a concentration-dependent manner. Conclusion: Resv inhibited cardiac hypertrophy indued by AngII, its mechanism might be through inhibiting the increases of protein content, surface area and the ERK-CREB signaling pathway.
