摘要:目的 探讨SD大鼠骨骼肌卫星细胞原代培养与鉴定的方法,以及大鼠脑胶质瘤细胞株体外培养并确定其生物学特性。 方法 取生后10周SD大鼠,在无菌条件下切取双侧腓肠肌与股中间肌,采用组织块原代和传代培养法和差速贴壁法培养与纯化骨骼肌卫星细胞,免疫荧光细胞化学予以鉴定。以及胰蛋白酶消化法进行大鼠脑胶质瘤细胞株体外传代培养。 结果 应用原代和分离纯化传至2~5代的快、慢骨骼肌卫星细胞,排除成纤维细胞,实施免疫细胞化学荧光抗体Desmin、 Fast-myosin、Slow-myosin分别予以鉴定。并采用大鼠脑胶质瘤细胞株体外传代培养,确定其生物学形态特征。 结论 本实验建立了骨骼肌卫星细胞原代培养、纯化和鉴定技术,以及大鼠脑胶质瘤细胞传代方法,为今后骨骼肌卫星干细胞与大鼠脑胶质瘤细胞深入研究奠定基础。?????

Preliminary Study of Skeletal muscle satellite cells and glioma cells cultured
Abstract:????? Objective :The SD rat skeletal muscle satellite cells were investigated in primary culture and identification methods, and rat glioma cell lines were cultured in vitro and determined for their biological properties. Methods :The bilateral gastrocnemius and vastus intermedius muscle in SD rats of 10 weeks after birth under sterile conditions were cut, using primary tissue culture and subculture, differential adherent culture and purification of skeletal muscle satellite cells, immunofluorescence cell chemical to be identified. And trypsin digestion method was implemented for rat glioma cell lines in vitro culture. Results :The fast and slow skeletal muscle satellite cells are applied in primary and purification spread 2-5 generations, fibroblasts excluded, the immunocytochemistry fluorescent antibody of Desmin, Fast-myosin, Slow-myosin was implemented to be separately identified. And the rat glioma cells in vitro culture were used to determine its biological morphology. Conclusion :The skeletal muscle satellite cells in primary culture, purification and identification technology were established. And rat glioma cell passage way for the future of skeletal muscle satellite stem cells and rat glioma cells in-depth study was established foundation.
