小麦成熟胚愈伤组织诱导及分化高效体系的建立

摘要:以L2,L3,L4,CN16四种小麦成熟胚为外植体,探究了不同激素浓度组合下小麦成熟胚脱分化与再分化情况。结果表明,一定浓度的2,4-D与KT配比有利于胚性愈伤组织的形成,4.0mg/L2,4-D+0.1mg/LKT+0.0mg/LNAA组合可作为最佳的诱导培养基选择;细胞分裂素的浓度影响愈伤组织再分化的效果,1.0~2.0mg/LNAA+0.1mg/LKT+0.5mg/L ZT+0.5mg/L6-BA比较适宜作为分化培养基的激素添加配比;受基因型影响,供试材料中L2的组培效果最差,L3,CN16最好,可选择L3,CN16作为建立小麦成熟胚愈伤组织诱导及分化高效培养体系的候选材料。

The Establishment of Efficient Culture System on Callus Induction and Differentiation from Wheat Mature Embryos
Abstract:Mature wheat embryos of four wheat varieties L2, L3, L4 and CN16 were cultured in MS medium with different combination of auxin and cytokinin. The results showed that the combination of 4.0mg/L 2,4-D+0.1mg/L KT+0.0mg/L NAA is beneficial to the formation of callus in induction medium. The effect of callus redifferentiation is regulated by different concentration of cytokinin, and 1.0~2.0mg/L NAA+0.1mg/L KT+0.5mg/L ZT+0.5mg/L 6-BA is more opportune in all differentiation medium. Because of the difference of genotypes,the mature embryos of wheat cultivar L2 displayed worse, instead, those of L3 and CN16 are better, L3 and CN16 can be candidate materials in an efficient system of mature wheat embryos cultured.
