离子辐射引起的细胞周期停滞模型

摘要:离子辐射是一种特殊的环境应激物,能够打断DNA的双链结构继而引起基因突变和癌症。目前国际上对于离子辐射的环境管理是基于线性无阈的剂量反应关系模型。可是实验数据显示J型的剂量反应关系占主导。细胞周期关卡停滞是离子辐射致癌的重要机理之一。为了模拟离子辐射的剂量反应关系,我们建立了离子辐射引起的细胞周期停滞模型,并与两步的癌症克隆增长模型相连接,模拟了离子辐射致癌的剂量反应关系。模型模拟结果显示了直线型到J型的剂量反应关系,鉴定其基本机理是由离子辐射引起的细胞周期关卡停滞时间趋于饱和而致。

Cell Cycle Checkpoint Arrest Model Induced by Ionizing Radiation
Abstract:Ionizing radiation is a special environmental stressor, it can cause double strand break and induce gene mutation and cancer. Currently a linear non threshold model is used for ionizing radiation regulation. But experimental data show that a J shaped dose response relationship is the dominating relationship. Cell cycle checkpoint arrest is an important mechanism for ionizing radiation induced cancer. In order to simulate the dose response relationship of ionizing radiation, we develop ionizing radiation induced cell cycle checkpoint arrest model and link it with two stage clonal growth cancer model. The model simulation results show a linear to J shaped dose response and identified its fundamental mechanism is that the cell cycle checkpoint arrest time tends to saturate with more doses.
