Csi-miR390、Csi-miR156超表达促进柑橘愈伤组织体细胞胚发生的潜力评估

摘要:为了验证Csi-MIR390和Csi-MIR156对柑橘体细胞胚发生能力的影响,从具有强胚性的伏令夏橙(Citrus sinensis)胚性愈伤组织中克隆两个miRNA的前体Csi-MIR390和Csi-MIR156,构建超量表达载体转化已丧失胚性的国庆一号温州蜜柑(Citrus unshiu)愈伤组织,分别获得超表达愈伤系3个和6个。经定量PCR检测,miR156的2个SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) 靶基因均不同程度下调表达;miR390只有一个功能未知的靶基因,但在超表达转化系与野生型或空载体转化系中没有差异。对这两个miRNA的超表达系愈伤组织进行生胚诱导,并未诱导出胚,但这两个miRNA超表达系生长速率均比野生型和空载体转化系慢,且透射电镜观察发现超表达系的细胞具有从愈伤组织一般的增殖状态向分化状态转变的趋势。此外,miR156的两个超表达系淀粉含量显著提高。以上结果说明miR390和miR156对柑橘体细胞胚发生有一定的作用。

Evaluation of somatic embryogenesis potential of citrus callus lines overexpressing Csi-miR390 and Csi-miR156
Abstract:In order to validate the potential role of Csi-MIR390 and Csi-MIR156 on citrus somatic embryogenesis, we cloned the precursors of Csi-MIR390 and Csi-MIR156 from embryogenic callus of Valencia sweet orange, which maintains strong embryogenesis capacity, and constructed overexpression vectors out of the precursors, which were then transformed to embryogenic callus of Satsuma mandarin 'Guoqing No.1' (Citrus unshiu) that had lost embryogenesis capacity. In two of the Csi-MIR156 overexpressing lines, the two SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) target transcription factors are down-regulated at different levels; while the only one target gene of csi-miR390, which is annotated as an 'unknown' gene, has no expression level difference in overexpressing lines, compared to wild type or empty vector transformants. Overexpressing Csi-MIR156 and Csi-MIR390 callus lines are unable to produce somatic embryos after being moved onto induction medium, however, they have slower growth rate relative to the wild type or empty vector transformants. By Transmission Electron Microscope (TEM) observation, we found that overexpressing Csi-MIR156 and Csi-MIR39 callus cells showed the tendency to differentiate, instead of the proliferation growth typical of callus; and the starch content in two of the Csi-MIR156 overexpressing callus lines were elevated significantly. These results suggest that miR390 and miR156 play certain roles in citrus somatic embryogenesis.
