摘要:目的:制备出一种高氧亲和力的血红蛋白氧载体。方法:以纯化的猪血红蛋白作为原料,用1-(3-二甲氨基丙基)-3-乙基碳二亚胺盐酸盐作为交联剂,并用N-羟基硫代琥珀酰亚胺加速聚合反应,将血红蛋白表面的羧基及胺基进行缩合形成肽键,从而使血红蛋白聚合在一起,形成聚合血红蛋白蛋白--即血红蛋白氧载体。结果:聚合血红蛋白的氧亲和力P50 = 6.60 torr,Hill系数n = 2.02,主要分子量分布从64 kDa - 1300 kDa。结论:以猪血红蛋白为原料合成出了一种高氧亲和力的血红蛋白氧载体。

Preparation of a high oxygen affinity hemoglobin-based oxygen carrier with porcine hemoglobin cross-linked by EDC
Abstract:A hemoglobin-based oxygen carrier (HBOC) with high oxygen affinity was prepared. Using the 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC)as the cross-linker and the hydroxy-2,5-dioxopyrrolidine-3-sulfonicacid sodium salt (Sulfo-NHS) as the catalyst, the pure porcine hemoglobin was polymerized and yielding a cross-linked derivative whose molecular weight distribute from 64 KDa to above 1300 KDa. From the oxygen equilibrium curves and Hill plots, the oxygen affinity (P50 = 6.60 torr) and Hill coefficient (n = 2.02) were obtained,the oxygen affinity was dramatic raised when compared with its native protein. ?????
