禽衣原体病喉头接种感染SPF鸡新模型的建立

摘要:本研究将鹦鹉热衣原体6BC株、CB7株、CB3株和HJ株分别通过滴鼻点眼和喉头接种感染15日龄SPF鸡,剂量为5×108.5TCID50/0.1mL,观察其临床症状和特征性的病理学变化,以探讨建立鹦鹉热衣原体SPF鸡模型。结果表明经喉头接种感染后,SPF鸡采食饮水量和体重显著性下降,四株鹦鹉热衣原体(6BC、CB7、CB3和HJ)感染后SPF鸡的平均体重相对增重率分别为30.2%、24.7%、13.8%、25.1%,健康对照组为46.5%。剖检后,感染后SPF鸡的肺脏肿大、出血、气囊炎、出血性肺炎、腹膜炎、心包炎和法氏囊局灶性出血或弥漫性出血。2种感染途径诱导的SPF鸡临床症状和病理学变化有差异,其中喉头接种病理学损伤高于滴鼻点眼途径。此外,本试验发现SPF鸡感染鹦鹉热衣原体上述菌株后,感染3d后未能检测出血清抗体,感染7d抗体水平显著增加,随着时间的推移,抗体水平开始下降。总之,本试验证实上述4株鹦鹉热衣原体以喉头接种方式可以诱导典型的鹦鹉热衣原体感染SPF鸡模型,且证明来源于鸭子CB7株和来源于肉鸽HJ分离株致病力性显著高于6BC株和CB3株。本试验首次证实鹦鹉热衣原体可以造成免疫功能的损伤。

A novel model for Avian Chlamydiosis by throat inoculation in SPF chickens
Abstract:Fifteen-day-old specific pathogen free chickens were given 5×108.5TCID50/0.1mL of four strains of Chlamydia psittaci by ocular-intranasal way and by throat injection, including 6BC, CB7, CB3 or HJ strains. Our current study aims to establish Chlamydia psittaci model in SPF chickens. Post inoculation by throat way, SPF chickens were observed to have lower feed take, and poor activity of drinking water as well as body weight gain. The average body-weight gain of infected chickens was 30.2%, 24.7%, 13.8% and 25.1%, respectively as compared to that of 46.5% in healthy group as the control. Moreover, the pathologies were characterized as fibrinous airsacculitis, pericarditis, peritonitis and pneumonia. More important, local hemorrhagic lesions were characterized in livers and bursa fabricius. In comparison with eye-intranasal route, more severe pathological lesions were observed in birds post inoculation by throat route. Furthermore, antibody was not detected on day 3, found to increase dramatically on day 7 and to decrease on day 10 post infection. From above data, 15-old-day SPF chickens displayed the typical lesions of avian Chlamydiosis by throat administration with above 4 strains while CB7 strain from ducks and HJ strain originated from pigeons are confirmed to be high pathogenic for SPF chickens. It is the first time that immune injury has been reported post inoculation with Chlamydia psittaci.
