砂梨离体植株几丁质酶基因的热处理诱导表达分析及克隆

摘要:热处理是果树病毒脱除中应用最广泛的一种方法,国内外学者大多集中于对病毒脱除效率的研究,而对热处理过程中寄主与病毒之间互作关系的研究甚少。本研究以前期构建的热处理诱导潜带苹果茎沟病毒(Apple stem grooving virus, ASGV)的砂梨离体植株的差减杂交文库中筛选获得的下调表达基因几丁质酶(EC)为研究对象,热处理和常温培养砂梨"黄花" (Pyrus pyrifolia) 离体植株为试验材料,采用Real-time PCR法分析了热处理进程中ASGV病毒含量变化与EC基因时间动态表达量之间的关系,根据所获的EST序列及NCBI登录的同源序列设计引物进而对EC基因进行了克隆和序列分析。结果表明,热处理1d 至10d 砂梨"黄花"植株中ASGV含量呈明显下降趋势,处理10d至35d无检测信号。热处理1d到35d EC基因表达量呈明显下降趋势,35d时达到最低值。EC基因序列全长为1053nt,编码334个aa,与已报道的来自砂梨"黄冠"分离物EC基因氨基酸序列一致率为86.9%。该研究结果为进一步研究EC基因蛋白功能提供材料,也为热处理条件下EC基因表达与病毒脱除之间的互作关系研究提供一定的参考信息。

Cloning and dynamic expression analysis of EC genes in Pyrus pyrifolia induced by thermotherapy
Abstract:Thermotherapy is used widely to eliminate viruses from fruit trees. Previous studies mainly focused on the efficiency of elimination viruses from hosts. However it is rarely understood to the interactions between hosts and virus during thermotherapy. In this study, the relationship of ASGV titers and the dynamic expression of chitinase (EC) screened from the reverse library in in vitro Pyrus pyrifolia "huanghua" plants during thermotherapy were further analyzed by RT-PCR and Real-time RT-PCR. The results indicated the expression quantity of EC gene were obviously decreased from 1d to 35d during thermotherapy and reached the lowest point at 35d, while the titers of ASGV in Pyrus pyrifolia decreased significantly from 1d to 10d, and no ASGV signal was detected from 10d to 35d. which will be helpful to know the relationship of EC dynamic expression titers and virus titers during the periods of heat treatment, and provide some information for further study the molecular mechanism of thermotherapy elimination viruses from pear plants. Full-length cDNA sequences of EC genes were cloned and analyzed. The results showed that the full-length cDNA sequence of 1053 bp from EC were obtained, which encoded 334 aa. The Blast search results showed the highest sequence identity of 86.9% at aa level of EC gene with that of Pyrus pyrifolia "huangguan" (GenBank accession No. FJ589784). The results provide materials for further study of EC gene protein function.
