PAK5与LMO2蛋白相互作用的初步研究

摘要:目的:应用酵母双杂交技术筛选PAK5(p21 activated kinase 5)的相互作用蛋白。方法:利用酵母双杂交技术,以PAK5为诱饵,从人胎脑文库中寻找出与PAK5相互作用的蛋白,并用GST pull-down,免疫共沉淀及共聚焦激光扫描显微镜技术进行验证。结果:经过酵母双杂交筛选,获得阳性克隆,进一步测序分析,确定筛选出的相互作用蛋白-LOM2(LIM domain Only protein 2)。结论:PAK5新的相互作用蛋白LMO2的发现,为揭示PAK5发挥生物学功能的分子机制提供线索,有望为肿瘤治疗提供新的理论依据。

Characterization of the novel interaction between PAK5 and LMO2
Abstract:Objective:Screening of novel PAK5-interacting proteins by yeast two-hybrid system.Methods:Using PAK5 as a bait,a human embryonal brain cDNA library was screened.After discarding the false postive clones,the truly positive clones were sequenced and submitted to BLAST analysis.The interaction was confirmed by in vitro pull-down assay,immunoprecipitation and laser scanning confocal microscopy.Results:LMO2,the novel PAK5-interacting protein were confirmed.Conclusion:This work would help to study the mechanism of the pleiotropic effects of PAK5,which might be a potential therapeutic strategy for tumor.
