家蚕乙酰胆碱酯酶和谷胱甘肽-S-转移酶的活性研究

摘要:乙酰胆碱酯酶和谷胱甘肽-S-转移酶活性与家蚕对农药的抗性密切相关。本文测定了对农药具有抗性及敏感性家蚕体内的乙酰胆碱酯酶和谷胱甘肽-S-转移酶活性。结果显示,农药抗性与敏感性家蚕中肠乙酰胆碱酯酶活性具有明显差异,同一时期比较,农药抗性家蚕中肠的AChE显著高于敏感性家蚕;抗性家蚕品种血液中谷胱甘肽-S-转移酶活性随着龄期的增加而显著升高,而敏感性蚕品种则维持较低的活性水平,到了5龄第6天,两者活性相差近5倍;抗性家蚕品种脂肪体谷胱甘肽-S-转移酶活性也明显高于敏感性蚕品种。以上研究表明有机磷和氨基甲酸酯杀虫剂农药进入蚕体后,中肠组织中的乙酰胆碱酯酶首先参与其解毒作用;当农药进入血液和脂肪体组织后谷胱甘肽-S-转移酶作为重要的解毒酶参与作用。

Studies of acetylcholinesterase and glutathione-S-transferase activities of Silkworm Bombyx mori L
Abstract:Acetylcholinesterase(AChE) and glutathione-S-transferase(GSTs) activities are closely related to silkworm's resistance to pesticides. In this paper we measured the AChE and GSTs activities in resistant and sensitive silkworm to pesticide. The results showed that AChE activity have significant differences in pesticide resistance and susceptibility silkworm midgut. In the same period of comparison, the AChE of pesticide-resistant silkworm midgut was significantly higher than the sensitive silkworm. In hemolymph of the pesticide-resistant silkworm, GSTs activity increased significantly with age development, while the sensitive silkworm remained lower activity levels. The difference between varieties was nearly five times in the sixth day of the fifth instar. In fat body of resistant silkworm GSTs activity was also significantly higher than the sensitive silkworm. The above study shows that when organophosphate and carbamate insecticides pesticides into the silkworm, AChE in the midgut do firstly participate in their detoxification, and when the pesticide enter the hemolymph and adipose tissue, GSTs act as an important enzyme involved in detoxification.
