雪松松针中2R,3R-二氢杨梅素的分离鉴定及其抗褐变作用的研究

摘要:本文的目的是研究雪松松针中主要抗褐变物质及其抗褐变作用。以酪氨酸酶抑制活性为筛选方法,采用大孔吸附树脂Diaion HP-20柱层析以及高效液相色谱法对雪松松针50%甲醇提取物进行分离纯化,并结合质谱与核磁共振数据鉴定化学结构。结果显示,其主要活性成分为2R,3R-二氢杨梅素。2R,3R-二氢杨梅素对单酚酶、多酚酶有较强的抑制作用,同时对ABTS自由基有较强的清除活性,并且呈现浓度依赖关系。鲜切苹果模型表明,2R,3R-二氢杨梅素对于防止苹果褐变有较好的延缓效果,并且与Vc有很好的协同效应。

Isolation and identification of 2R,3R-dihydromyricetin from pine needles of Cedrus deodara and its antibrowning effect
Abstract:The objective of the present study was to detect the main antibrowning compound(s) in pine needles of Cedrus deodara and its antibrowning effect. By bioassay guide of tyrosinase inhibitory activity, the main active compound was isolated and purified from 50% methanol extract of pine needles of C. deodara through macroporous resin Diaion HP-20 column chromatography and high performance liquid chromatography. Based on mass and NMR data, the active compound was identified as 2R,3R-dihydromyricetin, which showed the potent monophenolase and diphenolase inhibitory activities. Moreover, 2R,3R-dihydromyricetin had strong ABTS radical scavenging activity with a dose-dependent manner. In fresh-cut apple model, it was found that 2R,3R-dihydromyricetin was efficient in inhibiting browning of apple slices and showed a good synergistic effect with Vc.
