白芍总苷对糖尿病大鼠肾组织中内质网应激的影响

摘要:目的 探讨白芍总苷(TGP)对糖尿病肾组织中内质网应激的影响。方法 将链脲佐菌素(STZ)诱导的糖尿病大鼠模型随机分为模型组、TGP组(50、100、200mg/kgod灌胃),正常大鼠为对照组,8周后免疫组化法检测肾组织中GRP78、磷酸化的PERK(p-PERK)及磷酸化的eIF2α(p-eIF2α)的表达。结果 糖尿病组大鼠尿白蛋白排泄率(AER)明显高于对照组(P<0.01),TGP(50、100、200mg/kgod)给药8周后大鼠AER水平明显低于模型组(P<0.01)。模型组大鼠肾小球与肾小管-间质GRP78表达明显高于对照组(P<0.01),TGP(50、100、200mg/kgod)给药8周肾小球GRP78表达与模型组相比无明显差异,肾小管-间质GRP78表达明显低于模型组(P<0.01);各组大鼠肾小球未见p-PERK表达,模型组大鼠肾小管-间质p-PERK表达明显高于对照组(P<0.01),TGP(50、100、200mg/kgod)给药8周肾小管-间质p-PERK表达明显低于模型组(P<0.01);模型组大鼠肾小球与肾小管-间质p-eIF2α表达均明显高于对照组(P<0.01),TGP(50、100、200mg/kgod)给药8周肾小球及肾小管-间质p-eIF2α表达明显低于模型组(P<0.01,)。结论 糖尿病大鼠肾脏存在内质网应激反应,TGP抗糖尿病肾病作用可能与抑制内质网应激有关。

Effect of total glucosides of paeony on endoplasmic reticulum stress in the kidney from diabetic rats
Abstract: Objective To investigate the effect of Total glucosides of paeony (TGP) on endoplasmic reticulum stress in the kidney from experimental diabetic rats. Methods Diabetes was induced with streptozotocin in rats. Rats were randomly divided into three groups:control group, model group, model group treated with TGP. TGP 50,100,20mg/kg/d was orally administered once a day for 8 week.The expression of GRP78、p-PERK and p-eIF2α were determined by immunostaining in the kidney.Results Elevated 24h urinary albumin excretion rate was markedly decreased by TGP treatment with 50,100 and 200 mg/kg.d. Analysis of the immunostaining showed the expression of GRP78、p-PERK and p-eIF2α was significantly increased compared with control rats in the kidney.TGP treatment with 50,100 and 200 mg/kg.d could markedly reduced GRP78 and p-PERK expression in the tubulointerstitium,p-eIF2α expression in the glomeruli and tubulointerstitium.Conclusion The TGP protection of renal tissue in diabetic rats may be associated with inhibition of endoplasmic reticulum stress.
