槲皮素抑制3T3-L1脂肪细胞的分化

摘要:目的:明确槲皮素对3T3-L1前脂肪细胞成脂分化过程中的作用,探讨槲皮素抑制脂质沉积的作用机制。方法:本实验选取3T3-L1前脂肪细胞为靶细胞,将槲皮素加入其分化过程中,显微镜下观察,利用油红O染色确定其对脂肪化的作用,并利用定量PCR检测了槲皮素对脂肪化相关基因表达的影响。结果:20μM槲皮素可以降低3T3-L1细胞的脂肪化,减少脂质聚集,并且降低了3T3-L1细胞中脂肪化相关基因Pparγ、C/EBPα和Ap2的表达。结论:槲皮素可以降低3T3-L1脂肪细胞的分化。

Quercetin inhibits the differentiation of 3T3-L1 adipocytes
Abstract:Objective: To discover the effect of quercetin on differentiation of 3T3-L1 adipocyte and to explore the mechanism of inhibitory effect of quercetin on lipid deposition. Methods: Quercetin was added into the medium of 3T3-L1 during differentiation. After differentiation, the effect on lipid deposition was observed by microscope and measured by oil red O staining. In addition, the genes expression of 3T3-L1 differentiation were determined by quantitative RT-PCR. Results: 20 μM qercetin could suppress the differentiation of 3T3-L1 and lipid deposition and inhibit expressions of Pparγ、C/EBPα and Ap2. Conclusions: Quercetin decrease differentiation of 3T3-L1 adipocytes.
