难治性癫痫患者脑组织中RasGRF1启动子甲基化检测

摘要:目的:难治性癫痫发病机制尚不完全清楚,前期研究发现RasGRF1在难治性癫痫患者脑组织中明显减低,然而其表达调控机制尚不明确。本文研究难治性癫痫患者脑组织中RasGRF1启动子甲基化情况,从甲基化角度了解RasGRF1在难治性患者脑组织中表观调控作用及对下游p-ERK表达的影响。方法:收集40例难治性癫痫患者脑组织和30例对照组脑组织,采用甲基化特异性PCR (Methylation-Specific PCR,MS-PCR) 进行分析,比较两组启动子甲基化率是否存在差异;通过免疫印迹法比较病例组中RasGRF1甲基化、非甲基化以及对照组脑组织RasGRF1及下游p-ERK的表达。结果:在40难治性癫痫脑组织中,8例表现为RasGRF1启动子甲基化(27%),而在对照组中未检测到,病例组明显高于对照组 (p<0.05);RasGRF1在病例组中表达显著减低,其中在甲基化脑组织中最低;p-ERK1/2表达在对照组脑组织中显著低于病例组,其在病例组中的表达在RasGRF1甲基化脑组织较非甲基化样本低,但差异无显著性。结论:难治性癫痫患者脑组织中RasGRF1启动子甲基化明显增加,导致部分患者RasGRF1表达进一步降低,可能是机体一种反馈性保护机制。

Methylation of RasGRF1 in brain tissues of patients with refractory epilepsy
Abstract:Purpose: The mechanisms of refractory epilepsy have not completely understood yet. Previous study found that RasGRF1 significantly decreased in the brain of patients with intractable epilepsy, however, the regulatory mechanism of its expression has not yet clear. This paper focus on the methylation of the promoter of RasGRF1 in brain tissue of patients with epilepsy and its effect on the expression of downstream p-ERK. Methods: Methylation-specific PCR (MS-PCR) were used to analyze 30 samples of brain tissue of patients with intractable epilepsy and 20 samples of control group. Promoter methylation rates were compared. Expression of RasGRF1 and p-ERK were detected by immunoblotting. Comparison was conducted among RasGRF1 methylation group, the non methylation group of patient and control group. Results: Among 30 brain tissue samples of refractory epilepsy group, 8 samples showed RasGRF1 promoter methylation (27%), while no methylation was detected in the control group, the the difference was statistically significant (P<0.05). Expression of RasGRF1 in patient group was significantly decreased, which was the lowest in methylation group of patient; p-ERK1/2 expression was significantly lower in control group. Although the expression of p-ERK1/2 was lower in methylation group of patient, however, there was no significant difference compared with non methylation group of patient. Conclusion: RasGRF1 promoter methylation was significantly increased in brain tissues of patients with refractory epilepsy, and led to further reduced expression of RasGRF1 in part of patients, which may be a protective feedback mechanism of the brain.
