内源乳化法制备双歧杆菌微胶囊的研究

摘要:以海藻酸钠为壁材,采用内源乳化法制备两歧双歧杆菌微胶囊,用以提高其在人体胃肠液中的存活率。研究洗涤介质及制备工艺参数对两歧双歧杆菌微胶囊的粒径分布、包埋产率的影响规律,同时对微胶囊在模拟胃肠液中的耐受性和肠溶性进行评价。结果表明:在洗涤介质为1% (v/v)的Tween 80溶液,乳化剂Span 80添加量为油相体积的1% (v/v),水相和油相体积比为30:100,搅拌速度为400 r/min,碳酸钙和海藻酸钠的质量比为7.5:40,冰醋酸和碳酸钙的摩尔比为3.5:1时,制得的微胶囊平均粒径为116 μm,包埋产率为89.8%,与未微胶囊化的两歧双歧杆菌比较,微胶囊化使两歧双歧杆菌在模拟胃液和连续的胃肠液处理过程中的存活量分别提高了2个和3个对数值;经模拟肠液处理60 min,微胶囊全部崩解,具有良好的肠溶性。

Study on the microcapsules of Bifidobacterium prepared by internal emulsification method
Abstract:Bifidobacterium bifidum was encapsulated with alginate by internal emulsification method with the objective of enhancing viability during exposure to the adverse conditions of the gastro-intestinal tract. Effect of washing medium and the processing parameters on the characteristics of the microspheres containing Bifidobacterium bifidum were investigated. The acid tolerance and enteric solubility were also studied in this paper. Based on the size distribution, encapsulation efficiency, the optimal conditions were determined as follows: 1%(v/v)Tween 80 in aqueous solution as washing medium, the volume ratio of Span 80 to oil 1% (v/v),the volume ratio of water to oil 30:100, stirring speed 400 r/min,the mass ration of CaCO3 to alginate sodium 7.5:40,the mole ratio of glacial acetic acid to CaCO3 3.5:1. Under this condition, the average size of the microspheres reached to 116 μm , and the encapsulation yield of Bifidobacterium bifidum was 89.8%. Besides, compared to free cells, the viability of Bifidobacterium bifidum loaded in alginate during exposure to simulated gastric juice and sequential gastrio-intestinal juice increased two and three log respectively. After the treatment of stimulated intestinal juice for 60 min, almost all microcapsules disintegrated.
