热激蛋白(HSP20)对大肠杆菌耐受性的影响

摘要:目的:研究来自于腾冲嗜热菌(Thermoanaerobacter tengcongensis MB4)热激蛋白基因HSP20在大肠杆菌中过表达后对大肠杆菌耐受性的影响。方法:将gapA启动子和HSP20基因通过OE-PCR连接,转入大肠杆菌(Escherichia coli)获得重组工程菌 E. coli-HSP20。通过梯度升温发酵、添加有机溶剂的方法考察HSP20对大肠杆菌耐受性的影响。结果:过表达HSP20提高了大肠杆菌的耐热性和耐丁醇的功能将近1倍,乙醇和乙酸耐受性分别提高了9%和12.5%。结论:在高温、高浓度等胁迫环境下,腾冲嗜热菌HSP20促进大肠杆菌的生长,为工业发酵生产生物基产品的高效制造、节省成本提供了新思路。

Effect of heat shock protein (HSP20) on the stress resistance of Escherichia coli
Abstract:Objective:To study the effect of over-expression of heat shock protein (HSP20) from Thermoanaerobacter tengcongensis MB4 on the stress resistance of Escherichia coli. Method: A new genetic fragment gapA-HSP20 was made by combining Promoter gapA with HSP20 gene. Genetic fragment gapA-HSP20 was transformed into Escherichia coli to constructe engineered strain E. coli-HSP20. Investigate the stress resistance of engineered strain E. coli-HSP20 by gradient increasing temperature fermentation and adding organic solvent. Results: The heat-resisance and butanol resistance of E. coli was increased almost 1 fold under the over-expression of HSP20. Acetic acid and ethanol resistance was increased 9% and 12.5%. Conclusion: heat shock protein (HSP20) from Thermoanaerobacter tengcongensis MB4 promotes the growth of E.coli at high temperature and high concentration of organic sovent. These results provide a new idea for modern microorganisms industry.
