丹参多酚氧化酶基因的克隆及表达分析

摘要:为获得丹参中多酚氧化酶(PPO) 基因信息及其在诱导子处理后的表达情况,运用RACE技术从丹参(Salvia miltiorrhiza Bunge)毛状根中克隆到多酚氧化酶基因全长序列,其cDNA全长1930 bp,开放阅读框为1770 bp,编码589个氨基酸(GenBank登陆号为KF712274)。采用生物信息学分析发现该蛋白分子量约为65.8 kD,pI为6.56,具有典型的酪氨酸家族结构域,无信号肽结构,也没有跨膜螺旋,α-螺旋和β-折叠分散于整个多肽链中。在丹参毛状根培养液中加入不同诱导子进行处理,实时荧光定量PCR结果表明,该基因在酵母提取物处理中表达量显著上调,但在银离子、抗坏血酸和L-半胱氨酸处理中表达受到显著抑制。运用HPLC技术同步检测毛状根中丹酚酸B含量,显示出与基因表达相一致的趋势。上述结果说明丹参中多酚氧化酶基因对丹酚酸B合成具有正向调控作用。

Gene cloning of polyphenol oxidase from Salvia miltiorrhiza and its expression analysis
Abstract:In order to obtain the full-length sequence of polyphenol oxidase gene (PPO) and its expression in elicitor-treated hairy roots, it was cloned from hairy roots of Salvia miltiorrhiza Bunge by using RACE technique. The full length cDNA of PPO was 1930 bp with open reading frame 1770 bp, encoding a protein of 589 amino acids (GenBank No. KF712274). Bioinformatics analysis showed that the molecular weight of PPO gene was 65.8 kD with pI value 6.56, which had tryosinase family domain, but no trans-membrane helix or signal peptide. Both α-helix and β-sheet were found spreading in the whole polypeptide chain. Expression analysis of real-time PCR indicated that PPO transcript levels significantly increased in yeast extract (YE)-elicited hairy roots, whereas it significantly decreased in the other Ag+, ascorbic acid and cysteine treatments. In correspondingly, the accumulation of salvianolic acid B (SAB) was significantly increased in YE-treatment hairy roots, and was inhibited by the three elicitor treatments. These results suggest that PPO gene regulates the accumulation of SAB.
