淀粉样前体蛋白加工途径中分泌酶的研究进展

摘要:β-淀粉样肽(beta-amyloid peptide, Aβ) 的不溶性沉积物的积累是阿尔茨海默症的主要病理特征,其主要是β-淀粉样前体蛋白通过β-和γ-分泌酶顺序加工而产生,同时α-分泌酶可以在β-淀粉样肽内部切割从而阻止Aβ的产生。α-,β-和γ-分泌酶都对Aβ的产量起到了举足轻重的作用,所以对它们的结构与功能的深入理解有助于对阿尔兹海默症的预防及治疗。本文主要对β-淀粉样前体蛋白加工途径中的相关的分泌酶研究进展进行了系统综述。

Current progress on secretases in the processing of Amyloid Precursor Protein
Abstract:The accumulation of insoluble β-amyloid peptide sediment is the major hallmark of Alzheimer's disease. β-amyloid peptide is produced from the sequential processing of β-amyloid precursor protein by β-and γ-secretase, while α-secretase can process in the internal sequence of the β-amyloid peptide thereby preventing the generation of the β-amyloid peptid. α-,β-and γ-secretase have played a pivotal role in the processing of the β-amyloid precursor protein, so understanding of their structures and functions would facilitate the prevention and treatment of Alzheimer's disease. In this paper we review the current progressing of these three secretases.
