玉米ZmNBS105基因LRR区域的多态性和进化分析

摘要:核苷酸结合位点(NBS)类抗病基因是植物抗病基因中主要的一类,是近年来植物抗病基因研究的一大热点。本研究从玉米已鉴定的107个NBS类型基因中选取了ZmNBS105基因LRR区域作为研究对象,采用PCR方法从36个玉米自交系基因组中扩增测序后得到30个自交系的有效测序结果。利用生物信息学方法分析了序列的相似性、多态性位点及核酸多态性等参数,研究结果表明ZmNBS105基因LRR区域在DNA水平上变异程度很低,核苷酸多态性π值为0.00096,远小于其它已知抗病基因。从进化树聚类图可以看出,进化树明显分为两枝,且分支内序列差异非常小,表明该段序列非常保守。同时,该区域的Ka/Ks比值小于1,说明该基因LRR 区域在进化过程中受到纯化选择作用。本研究为分析NBS类型抗病基因的分子进化机制提供了理论依据,也为深入研究其功能奠定了基础。

Nucleotide diversity and evolution analysis of LRR region at ZmNBS105 locus in maize
Abstract:Nucleotide-binding site (NBS) disease resistance gene is a major class in plant disease resistance genes, which becomes a focus in current studies of plant disease resistance. In this study, We choose ZmNBS105 gene as a research object from 107 genes with NBS domain that were identified in maize, and the LRR region of the ZmNBS105 were amplified with PCR method from 36 maize inbred lines. Finally, 30 effective sequences were obtained. Then, the similarities, polymorphic sites, and nucleotide diversities of the sequences were estimated by bioinformatics methods. The results showed that low nucleotide polymorphism (0.00096) existed in the LRR region of ZmNBS105 gene, which is much less than other known resistance genes. The phylogenetic tree revealed that the 30 sequences can divided into two branches, and the sequences of each branch are similar, indicating that these sequences are highly conserved. Meanwhile, the Ka/Ks ratio of the region is less than 1, suggesting that the LRR region of ZmNBS105 is subjected to purifying selection during the evolutionary process. Our study provides a theoretical basis for analyzing the molecular evolution mechanism of NBS disease resistance gene and also lays a foundation for further study of the function.
