金纳米粒子与胃蛋白酶相互作用的研究

摘要:利用荧光光谱、同步荧光光谱和紫外可见吸收光谱,在生理条件下研究了纳米金与胃蛋白酶之间的相互作用机制。研究结果表明,纳米金对胃蛋白酶的荧光具有明显的猝灭作用,它们之间相互作用的机理主要是一个形成了复合物的静态猝灭过程。通过对实验数据的处理,可以计算出不同温度下的猝灭常数、结合常数、结合位点数以及相应的热力学函数。同时,由计算得到的热力学函数△H 、△S 和△G可以推断出氢键和范德华力是相互作用过程的主要作用力。此外,运用同步荧光光谱和紫外可见吸收光谱探讨了纳米金对胃蛋白酶构象的影响。

The Interaction Mechanism of Gold Nanoparticles with Pepsin
Abstract:Under physiological conditions, the interaction mechanism of gold nanoparticles with pepsin was investigated by fluorescence, synchronous fluorescence and UV-Vis absorption spectroscopic techniques. The experimental results indicate that gold nanoparticles have a strong ability to quench the intrinsic fluorescence of pepsin through the static quenching procedure with the formation of a ground state complex. Based on the experimental data, the quenching constants, the binding constant, the binding sites and the thermodynamic parameters were evaluated in different temperatures. Additionally, according to the calculated thermodynamic parameters, it can be concluded that hydrogen bonding and van der Waals forces may play an important role in the binding process. Furthermore, the effects of gold nanoparticles on pepsin conformation were investigated by synchronous spectroscopy and UV-Vis absorption spectroscopy.
