重离子束和X射线辐照对舌鳞癌细胞Tca8113中MMP-2,MMP-9表达的影响

摘要:目的 探讨两种不同LET射线对舌鳞癌细胞Tca8113中MMP-2和MMP-9表达含量的影响。 方法:采用不同剂量的重离子束以及X射线对舌鳞癌细胞Tca8113进行辐照,用Western-blot检测Tca8113在辐照后不同时间点上MMP-2和MMP-9表达含量的变化。 结果:1Gy X射线在辐照后12h、24h和48h,MMP-2的表达含量均有所上调(p<0.05),其表达含量于24h时达到顶峰(p<0.01);4Gy X射线辐照后的细胞中MMP-2在各个时间点的表达水平虽有所减少,但并不具备统计学差异(p>0.05)。不同剂量的X射线均对细胞中MMP-9的表达没有明显影响(p>0.05)。1Gy重离子束对舌鳞癌细胞进行辐照后,MMP-2于12h时出现表达含量的升高,但其表达水平于24h和48h时又出现了明显下降(p<0.05);而相同剂量下重离子束对舌鳞癌细胞中MMP-9的表达没有明显影响(p>0.05)。在使用4Gy重离子束对肿瘤细胞进行辐照后,MMP-2和MMP-9的表达均有明显降低(p<0.05)。 结论:在相同的辐照剂量下,重离子束在抑制肿瘤体外侵袭能力方面比X射线有更高的相对生物学效率。

The expression of MMP-2, MMP-9 in tongue squamous cell carcinoma cell Tca8113 irradiated by heavy ion beam and X-ray
Abstract:Objective: To investigate the expression of MMP-2 and MMP-9 in tongue squamous cell carcinoma cell irradiated by different LET irradiation. Method: Tca8113 was irradiated by different doses of heavy ion beam and X-ray. The expression of MMP-2 and MMP-9 were measured in different time-points by Western-blot. Result: The contents of MMP-2 irradiated by 1Gy X-ray all increase after 12 hours, 24 hours and 48 hours (p <0.05). The expression content peak appears at the point of 24 hours (p<0.01). The expression level of MMP-2 in the cell decreases at different time-points after 4Gy X-ray radiation but it bears no statistical differences (p>0.05). After conducting irradiation on 1Gy and 4Gy X-ray, the contents of MMP-9 in cells doesn't appear any obvious change (p >0.05).After the irradiation of 1Gy heavy ion beam towards squamous cell carcinoma of the tongue, the MMP-2 shows the increase of expression content at the point of 12 hours while its expression level decreases obviously at the time points of 24 hours and 48 hours (p<0.05). Low-dose heavy ion beam doesn't have obvious effects on the expression of MMP-9 in tongue squamous carcinoma cells (p>0.05); after using 4Gy heavy ion beam to irradiate the tumor cells, the expressions of MMP-2 and MMP-9 have obvious decreases (p<0.05). Conclusion: Compared with X-rays, heavy ion beams have higher biology efficiency in tumor invasion suppression.
